Crikey! Now here is something that I can write about!
My personal opinion is that there is no best game of all time, but I can make genre-specific arguments for certain games.
Personally, I think that for this, games should be considered within the context of the time in which they came out.
FPS:
I'm going to have to say Doom on this one. Let's face it, as far as PC FPS games go, Doom was absolutely huge in sales and in impact on the genre, and while there have been many successful first-person shooters, Doom is patriarch of FPS (yes, I know about Wolfenstein 3d, and it was a good game. However, it didn't catch on nearly so much as Doom did).
RPG:
This is a tough one. I've played many great RPGs, and I don't have any particular one that dominates the others in context. My votes would go to Final Fantasy 6, Chrono Trigger, Earthbound, and Fallout 2. Personally, I think Fallout 2 and Earthbound were the best of those, but they didn't really have the widespread impact on RPGs that FF6 and CT had.
RTS:
Ah, my favorite genre! In terms of truly great RTS games, I would have to go with Command and Conquer:Red Alert, StarCraft, and Total Annihilation. The first is fairly well-known, it was basically the C&C game in which Westwood did pretty much everything right. The second is fucking StarCraft! Need I say more? Lastly, Total Annihilation makes it to the list of amazing fucking games that somehow disappeared without a trace. It was destruction on a truly epic scale, and had it not come out at precisely the same time as StarCraft (or if it had better marketing), it could have become a monolithic gaming franchise.
MMORPG:
I don't play MMORPGs, so I can't really comment on them.
Fighting games:
Street Fighter 2. Humongous impact on fighting games, and spawned stupid numbers of sequels.
Platform Side-Scroller:
Super Mario Brothers 3. They say it's the greatest game of all time, and while I disagree with that, I will agree that it's the greatest one in this category. Honorable mentions go to Megaman 2 and Duke Nukem 2.
Adventure Game:
A nearly dead genre, but there were moments of absolute brilliance in some of these games. I've not played a lot of them, but my personal favorite was Sam and Max Hit the Road. It was a fun, hilarious game that never ceased to amaze me in how ridiculous it was.
Miscellaneous:
Sim City 2000. A fantastic game, if you'd never played the sequels, you'd be blown away at how well done it is.
Those are my opinions on the subject. I'm biased toward the games that I played while growing up, though, possibly to the exclusion of recent games :/ _________________ The official MILF-masta of the AlP forums.

FPS: HL2... it just roxorz...
RPG: Fallout and Fallout 2. If anyone disagrees I'll shove so much Mentats down you're throat you'll lose your other stats caus your Int is getting so high... j/k
RTS: Dune 2000. C&C RA2 was also good, but Dune 2000 was better I think... if you look at the atmosphere of it all... and the awesome videos etc etc...
MMORPG: Well, I played Lineage 2 a while and it was cool... for the rest, I could really recommend KnightOnline, Shattered Galaxy and Runescape... well, Runescape if you don't have a high-end pc.
Fighting games: Fighting games for the pc... can't really think of anything... well... Red vs Blue is ofcourse awesomeness perfected... twice...
Platform Side-Scroller: Duke Nukem 2 was fun... so was Abuse...
Adventure Game: Tough one... I once played something where you had to break in a house and solve numerous puzzles that had to do with locking mechanisms...
Miscellaneous: Evolva... must be mentioned. That game rocks. If you don't know what it is, read this. I really think it rocks... _________________

You fight like a dairy farmer!
Aramor mentioning Evolva reminded me of all those great commercial flops, so here are a few of my favourites that inexplicably failed to sell well: Outcast, Thief series, Startopia, and oh lets say Urban Chaos. _________________

I just realized that my answer contains console games, when the question was specifically for PC games... so just ignore the console titles :/ _________________ The official MILF-masta of the AlP forums.

I never got to play that one (Day of the Tentacle), though I have only heard good things about it. I also liked Grim Fandango.

Supreme Commander is Total Annihilation 2 in all but name, it comes out next year. Must agree that Baldur's Gate 2 is pretty much perfect. _________________

I'm going to have to include some platform since FPS and MMORPGs are the only PC games I play
MMORPG: World of Warcraft and Star Wars Galaxies are a tie for me.
RPG: I know it's old, but I'm going to have to stick with my all time favorite: Final Fantasy III (US, SNES) and FF7 (PS1) is a close second. Vampire: Bloodlines for the PC, partially an RPG, is up there too.
FPS: I still love Battlefield 1942.
RTS: The first game I ever really got into in this category may not be the best in the graphics and options department, but the first Command and Conquer holds a special place in my heart.
Stragety & Map Conquest: Civilization 3 and Romance of the Three Kingdoms 8 are dead even.
Fighting: Street Fighter 3, there will never be a better fighting game, ever, never, ever. _________________ All I want, to know, all I want, with just a touch of my burning hand I send my astro zombies to rape the land. Prime directive, exterminate the whole human race! |

The only PC games I play are exploration games like Myst and Zork, and text adventures. Zork Zero rules all. Also, the villain in Myst 3: Exile was the crazy scientist in Alien Resurrection. Just a fun fact.
Oh, and Baldur's Gate 2 is really good. _________________ The depressing thing about tennis is that no matter how good I get, I'll never be as good as a wall.
